How many five-letter words use five different letters
Rather more than people expect. A clear majority of five-letter English words contain no repeated letter at all, which makes the repeated-letter words the exception rather than the rule, and explains why they are so easy to overlook.

What makes this set structurally different
Words without repeats have to distribute their consonants and vowels more evenly, because there is no way to lean on a doubled letter to fill a slot. That pushes them towards the consonant-vowel-consonant-vowel-consonant shape and its close variants.
The practical consequence is that this list is unusually rich in words with two separated vowels, and unusually poor in the -LL, -SS and -TT endings that account for a large share of English five-letter words generally.
Why the distinction is useful
If you are filling a five-letter slot and you know two of the letters are the same, this list is precisely the set to exclude. If you know all five are different, it is the set to search. Either way, splitting five-letter words along this line cuts the search space roughly in half at no cost.
It is also the right list when you want a word that tests as many distinct letters as possible, such as a guess in a clue-based puzzle, or an opening play that touches the most new tiles.
Letters that resist appearing here
A few letters are underrepresented for structural reasons. S is common in five-letter words largely as a plural ending, and plurals of four-letter words frequently repeat a letter from the stem. Words built on double consonants drop out of this list entirely.
Q is nearly absent for a different reason: it almost always drags a U with it, which consumes two of five slots and leaves little room to place three more distinct letters that form a word.
